Research Support - Laboratory/Non-Laboratory, Staff/Administrative
The Division of Rheumatic and Autoimmune Diseases in the Department of Medicine is seeking a Researcher 1 to join the team. Our lab is interested in understanding the fundamental biological mechanisms for immunological tolerance during autoimmunity, cancer immunology and transplantation, directed at therapeutic intervention strategies. We are using autoimmune animal models to understand biological processes involved with autoimmune diabetes and cancer treatments.
Duties and Responsibilities: The post-holder will be perform molecular biology cloning of new antibodies, new peptide:MHC monomers, and characterize these reagents using biochemistry, western blotting, flow cytometry as assigned by the PI. This will include the generation of novel antibodies for therapy, purification of protein reagents for unique molecules to identify rare T and B cell populations. This post-holder will assist in normal lab and mouse colony maintenance and perform other duties as assigned by the PI when the lab manager is unavailable. • Generation of novel antibodies for therapy, purification of protein reagents for unique molecules to identify rare T and B cell populations, molecular cloning, sequencing, western blotting, immunoprecipitation, protein purification, and bioreactor protein production will be main duties. Assistance to maintain unique transgenic and knockout stocks and strains of animals; operations and records and interacting and collaborating with research staff; will participate in monitoring rodent populations by PCR, Flow Cytometry, and diabetes assessment prior to and following treatments. (50%) • Perform research projects according to established protocols and following direction of PI to include: cell culture, drug preparation, tolerance experiments, diabetes assessment, plasmid transfection, drug preparation and administration, assist with PBMC and tissue preparations, assist with surgical procedures, and perform flow cytometry analysis, immunohistochemistry, and cellular imaging 2-photon microscopy (10%) • Lab organization, lab cleaning, records keeping, solutions and equipment maintenance (30%) • Data collection and analysis. Prepare written and graphical descriptions of results to discuss with PI. Review scientific literature and technical documents. (10%)
All required qualifications must be documented on application materials.
Required Qualifications: • B.S. or B.A. in biological sciences or related field, or education and work experience in biological science to equal a combination of 4 years. • Must have basic experience working with laboratory mice. • The applicant must be able to interact professionally with all faculty, students and staff who utilize the facility. • Required computer skills include word-processing, protein analysis, DNA blast searches, spreadsheets, database and presentation, preferably with Microsoft Office Software. • Excellent organizational skills, communication, and attention to detail
Preferred Qualifications: • Preferred education and/or work experience in biochemistry. • Preferred basic understanding of immunology through course work. • Preferred basic experience working with sterile tissue culture conditions for insect or mammalian cell lines as a laboratory course or research experience in a lab setting. • Preferred basic experience working with mice including handling mice in specific pathogen free (SPF) conditions, breeding, weaning, and sample collection. • Preferred basic ability to harvest mouse tissues under sterile cell culture techniques/conditions. • Preferred basic laboratory experience using flow cytometry. • Must have 1 year laboratory technical experience in molecular biology and cloning in a research laboratory setting or a passing grade for molecular biology. • Must have 1 year of laboratory technical experience performing protein isolation, DNA sequencing, immunoprecipitation and western blots or a passing grade for a course in biochemistry.
