Rutgers, New Jersey Medical School is seeking a Postdoctoral Fellow for Pharmacology, Physiology and Neuroscience. Under the direction of the Principal Investigator (PI), the Postdoctoral Fellow will (1) First, determine the effect of differentiation and cell fate on intestinal tight junction (TJ) composition and macromolecular permeability. Study 1.1.1 will analyze the composition and localization of major TJ proteins in ISCs, ENT, GOB, PAN as well as EEC cells, and then Study 1.1.2 will determine the flux of different-sized dextrans in enteroids enriched in different cell types, to estimate cell-type induced differences in macromolecular permeability as well in steric properties of the leak pathway. Study 1.2 will examine whether, upon completion of differentiation, the TJ composition and macromolecular permeability of postmitotic cells are mutable or permanent conditions, by determining whether forced dedifferentiation of postmitotic, fully differentiated absorptive enterocytes and secretory GOB or PAN cells results in reacquisition of TJ functions characteristic of ISCs. Study 1.3 will examine whether an absorptive or secretory cell fate affects the plasticity of TJs and the capacity for dedifferentiation. (2) determine the mechanistic contributions of major TJ proteins thought to regulate the leak pathway to cell type-dependent as well as differentiation-related changes in TJ composition and macromolecular permeability. In Study 2.1, TJ composition and macromolecular flux will be determined in ISC, ENT, GOB, PAN and EEC enteroids obtained from mouse models unable to express occludin. Study 2.2 and 2.3 will examine TJ characteristics of enteroids enriched in different cell types, in mouse models without tricellulin and ZO-1, respectively.
Among the key duties of this position are the following:
Participates in experimental design; breeds and maintains mouse genetic models; and carries out experiments to meet, at minimum, the specific aims above, as well as related laboratory objectives.
Participates in meetings with project PI and his collaborators, presents in laboratory meetings, coordinates activities with other postdoctoral fellow.
Operates and maintains experimental equipment.
Records and analyzes data from experiments.
Maintains a proper data notebook pursuant to the standards of the Department and the PI.
Contributes to the intellectual development of the project, and the writing of manuscripts.
Submit proposals for post-doctoral fellowship funding.
Supervises graduate, undergraduate and high school students in the laboratory.
Performs other related duties as assigned.
Minimum Education and Experience:
MD/PhD or Ph.D. in microbiology or related disciplines.
Experience working with gut microbiome, metabolomics, bioinformatics and statistical methods in microbiome research essential.
Demonstrated experience in gnotobiotic mice models, FACS and confocal microscopy is desirable.
Individual must be able to work independently and work with a large multidisciplinary research team.
Excellent verbal communication and writing skills in English are a precondition.
